延胡索酸酶[3](英语:Fumarase,亦称为延胡索酸水合酶,EC 4.2.1.2)属于柠檬酸循环中的一种酶,主要催化延胡索酸水和形成L-苹果酸。该酶的催化反应是可逆的,但具有严格的立体选择性,水裂解的羟基被严格地加到延胡索酸分子双键的一侧,而氢被加到另一侧。故正反应中只催化反式双键(延胡索酸)的水化,而不能催化马来酸, 而逆反应只催化形成苹果酸的L-异构体。
