FokI是一种存在于细菌Flavobacterium okeanokoites的type IIS限制酶,含有位于N端DNA结合区块(N-terminal DNA-binding domain),以及一个位于C端的非专一性DNA切割区块[2]。当此酵素与DNA辨识并结合于5'-GGATG-3'识别位点时,将会把位于结合位置下游的9到13个核苷酸切除(不需特定序列)[3]FokI的分子量为65.4 kDA,含有587个氨基酸
