NCI-H295R细胞

NCI-H295R(也称为H295R)是一种会对血管紧张素II及离子反应,并且会产生类固醇人类肾上腺皮质腺癌细胞[1],最初于1980年从一名诊断为肾上腺皮质癌英语Adrenocortical carcinoma的48岁女性患者中分离出来[2],该细胞系称为NCI-H295。由于NCI-H295细胞株的生长速度慢且易于分离,因此会选择具有更好的单层附著性和更快生长的细胞群。同时NCI-H295细胞经改变培养条件而获得三种细胞株,分别称为H295R-S1H295R-S2H295R-S3[3]。这三种细胞株均以单层的粘附培养物方式生长[1]

H295R细胞常用于对人肾上腺皮质生长和功能英语Function_(biology)研究[3],因为其在激素分泌和细胞特征上与原代培养的人肾上腺皮质细胞最接近。有学者指出H295R细胞系是一种有效的体外筛选环境类固醇激素干扰物及研究其机制的细胞模型[4],能够从基因表达、活性度激素水平三个层面进行检测[5],并且能够评价环境中的污染物对细胞活性和细胞毒性的影响,继而区别化学物是通过直接的细胞毒性作用的影响产生效应,还是间接的生成类固醇通路的影响产生效应。美国国家环境保护署也利用H295R细胞系研究环境中污染物的内分泌干扰效应[6]
